Restriction digests of the clone give the following sizes (kb): BamHI--20.0, 15.0, 9.0; EcoRI--20.0, 15.0, 9.0; EcoRI/PvuII--17.0, 10.0, 4.6, 4.3, 3.9, 1.8, 0.6; HindIII--40.0, 4.6; PstI--17.0, 7.0, 4.6, 2.8, 2.5, 2.0, 1.8, 1.5, 1.2, 1.15, 0.85, 0.55, 0.3; SalI--20.0, 9.0, 5.0 (triplet). The sequenced portion also contains a PvuII, a HindIII and an EcoRI site. This clone was isolated from an EMBL4 library, grown on E. coli LE392 and characterized at the Cytogenetics and Cancer Genetics laboratory of Memorial Sloan Kettering Cancer Center, New York. Date of isolation - Sept.-Oct. 1986. A 2.54 kb PstI fragment from the insert has been sequenced (X12660) and contains a rearranged Smu-Cmu region. The sequenced fragment contains 26 direct repeats 10-44 bp long, 51 inverted repeats, and more than 20 putative Ig recombinational signals. |
