Digestion with BamHI yields 2 vector fragments (5.4, 5.2 kb) and the 2.3 kb insert (+ some pcD). Restriction digests of the clone give the following sizes (kb): NotI--uncut; BssHII--13.0; SfiI--7.6, 5.4; PstI--7.4, 3.9, 1.5; SacI--13.0; BamHI--5.2 doublet, 2.3; HindIII--13.0; BglII--13.0. Vector contains EBV oriP and EBNA-1 to ensure replication in absence of EBNA. Maintained as episomes as 2-10 copies per lymphoblastoid cell; stable as long as selection is applied. J107 cells were transformed with DNA from the T-lymphoma JM cell line. DNA from secondary transfectants was used to isolate the clone. |
